Q: does it charge by usb and how long will it stay charged?

The USB cable is for power. There is not a fixed battery, however there is an option to use with xAAA batteries.

Q: Can you remove the rows?

Not without damage. They are not meant to be removed. If you are looking for a flat backlit surface, look for a light box.

Q: How many of each letter are supplied with the light box? What if I need 3 of the letter "S"?

My granddaughter (whom I bought it for)… said there are 4 of each letter.

Q: Is there a timer function on this ? Will it turn off after a set time?

There is no timer function. It stays on until you physically turn it off.
