Q: Should the primer be up or down?Does it matter?

Depends. I use different sizes of the boxes for different ammo at the range. Primer up for pistol ammo allows you to pull the shells by grasping the head of the shell with the finger tips (easier than grabbing a tapered bullet tip in a confined space). Rifle rounds almost have to be primer down, as longer rounds balanced on their tip tend to flop around in the box.

Q: Will they hold .45 acp and .50 ae?

It will fit .45 ACP but the rounds will drop down too low to pick out. Don't know about .50 AE.

No it is not the item is far too small. review the TSA regulations if you travel by air and driving it must be in a separate again. a lockable gun box in addition to your weapon should work. Review or ask the federal and state personnel to see if your containers will comply I've found TSA surprisingly helpful as with the LEOs in the jurisdictions of travel.
