Would the sugar skull hose fit someone who is a plus size 22??

They may, but they will probably not be comfortable. I am a 14/16 and the material is really strong which translates to tighter fit. If you're wearing for a photoshoot, or dinner and a movie, I think you will be ok.

Yes, pull them up like regular hose

I weigh around 160 something and i have thick thighs and a large behind. Will the one size fit me ?

My hips are 55inches around and they fit good. So I say yes because I weigh around 250 and have a pear shape.
